Biodegradable plastics offer a promising alternative for petroleum based synthetic plastics. The synthetic plastic products emit green house gases like carbon dioxide on degradation and take up a lot of space in landfills. On the other hand, biodegradable plastics can be easily synthesized in bacteria or plants in natural conditions. They also can be disposed off and degraded easily. The degradation process of biodegradable plastics releases substances that are less damaging to the environment when compared to that of the synthetic plastics thereby reducing the pollution. • Centrifuge at 8,000rpm for 20mins. • Discard supernatant and wash pellet with 10ml diethyl ether. • Centrifuge at 8,000rpm for 10mins. • Pellet dried at 1050C for 24hrs to get PHB crystals. 4.5.4.2 Sodium hypochlorite- Chloroform Method • Centrifuge culture at 8,000rpm for 20mins in prewashed tubes with chloroform and ethanol. • Discard the supernatant. • Pellet was treated with 5ml Sodium Hypochlorite and incubated at 300C for 2hrs. • Centrifuge at 5,000rpm for 15mins. • Discard supernatant and add 5ml distilled water to the pellet and centrifuge at 5,000rpm for 15mins. • Discard supernatant and add 5ml of Acetone to the pellet and centrifuge at 5,000rpm for 15mins. • Discard supernatant and add 5ml of Methanol to the pellet and centrifuge at 5,000rpm for 15mins. • Discard supernatant and dissolve pellet in 5ml of boiling chloroform. • Evaporate chloroform by pouring the solution in glass tray and kept at 40C.
