The Rose Bengal Plate Test (RBPT) antigen from Brucella melitensis local isolates were developed in this study. The performance of the assay was investigated using serum samples collected from goats. A total of 1063 serum samples obtained from goats were examined for the presence of antibodies against Brucella by in-house RBPT (LRBPT), commercial RBPT (Veterinary Laboratory Agency –VLA, UK) and Compliment Fixation test (CFT).The sensitivity and specificity was calculated using CFT as the gold standard. Out of 1063 goats sera analyzed 364 (34.24%), 335 (31.51%), and 373 (35.08%) were found to be positive by LRBPT, commercial RBPT, and CFT, respectively . The sensitivity calculated for the LRBPT, was 90.1% compared to commercial RBPT 85.0%. However, the specificity of the LRBPT was lower (95.9%), than the commercial RBPT (97.4%). …show more content…

However, the PPV of the in-house rose Bengal plate test (92.3%) is lower than the PPV of the commercial rose Bengal plate test (94.6%). In addition, the NPV of the in- house RPBT (94.7%) is higher than the NPV of the cRPBT
Mannitol Salt Agar (MSA) plate, MacConkey agar (MC) plate, Eosin Methylene Blue agar (EMB), and Hektoen Enteric Agar (HEA) (3). The MacConkey agar plate and the Mannitol Salt agar plate are both used in the identification of the unknown. The MC plate is a selective and differential medium. It is considered a selective medium because the bile salts and crystal violet aspect of the medium prevent the growth of gram positive bacteria (3). This medium is differential because of the lactose and neutral red.

In the summer of 2015, a pink-eye (conjunctivitis) outbreak occurred on the family ranch. This was the greatest challenge I have faced in my beef production SAE. Many of the family cattle as well as many of my own cattle were infected with the contagious bacteria or virus. The outbreak was spreading quickly and if left untreated would blind infected animals, infected animals would also not be able to be sold. The correct vaccine would need to be found through research and administered to infected animals.

After a gram stain was done unknown #257 was identified as a gram positive organism because when observed under the microscope the organism appeared purple with cocci in clusters. The organism was also catalase positive which means that it produced enzyme catalase and bubbled when hydrogen peroxide was added to it. Three test were conducted based on the result of the gram staining procedure. Blood agar with a Novobiocin disk was chosen as well as DNase (DNA) and Mannitol Salts (MSA) agar. The Blood agar is a bright red, opaque plate and the streaking or the inoculation technique was a modified streaking for isolation with a heavy quadrant one.

Dr. Sluis-Cremer’s accom-plishments as a biochemist, translational scientist, and educator are exceptional and most deserving of promotion to this rank. Dr. Sluis-Cremer’s academic training and record of research, teaching, mentoring and service are thoroughly reviewed below. Training and Faculty Record Dr. Sluis-Cremer received his Ph.D. in Biophysics and Biochemistry in 1997 from the University of Witwatersrand, Johannesburg, South Africa. From 1998 to 2001, he completed a Postdoctoral Fellowship with Michael A. Parniak, Ph.D., at the McGill University AIDS Center, Lady Davis Institute for Medical Research, Jewish General Hospital, Montreal, Canada.

Results Double transfection do not change Ld1S parasites’ performance The double transfection approach generated bioluminescent and fluorescent Ld1S parasites as virulent as Ld1S wild-type ones. Indeed, the virulence of the transfected parasites was verified at each stage of the process.
