The carbon balance analysis would determine the carbon source transformation along the biofiltration process. The analysis provided details how toluene carbon distributed into products of biomass accumulation in the filter bed, CO2 or mineralization, and leachate partition during the biofilter process.
Regarding to calculation for carbon weight, daily toluene removal rate, wet biomass, CO2 generated concentration, and TC in leachate were performed through following responsible equations in Table 2.2 below.

These slides were in order to grow biofilm for 2 weeks. Also, another reactor biofilter system, called BF3, was set up provided with all the air feeding line connection with air pump, customized toluene generation bottle, UV lamp, and spray unit.
After 2 weeks of growing biofilms, glass slides were retrieved from both biofilters and placed inside BF3. Then, the ozone concentration was varied between 200-800 mg/m3, and those slides were treated with varied period was between 5 min to 24 hours. More activated copper was added to the filtrate and vacuum filtration was perform again. The error arrived when the filter paper was not replaced because there might have been a hole inside the paper that prevents all the activated carbon to be filter. Since the paper was pitch black, it was impossible to see if there is leakage. The filtrate was less blue after the second filtration. A third vacuum filtration was performed after adding much more activated carbon.
Identification of bacteria within Unknown Culture #21 In this experiment, an unknown culture of two different types of bacteria was assigned to each person, a number of tests were performed to isolate and identify these bacterial cells. Based on knowledge from the previous experiments completed in lab, a basic understanding of each type of bacteria was used to create a flow chart that would aid the process of identifying the unknown bacteria within the culture. A gram stain that is performed initially will narrow down the types of tests certain bacteria will and will not respond to. In addition to the gram stain, some of the tests that were used include, a catalase test, an Eosin methylene blue (EMB) agar test, a bile esculin test, and a 6.5% sodium chloride (NaCl) test.
Mannitol Salt Agar (MSA) plate, MacConkey agar (MC) plate, Eosin Methylene Blue agar (EMB), and Hektoen Enteric Agar (HEA) (3). It is considered a selective medium because the bile salts and crystal violet aspect of the medium prevent the growth of gram positive bacteria (3). 1. Introduction 1.1 History of the Gowanus Canal The Gowanus Canal, located in the New York City borough of Brooklyn, is one of the most heavily contaminated bodies of water in the United States. The dimensions of the Gowanus canal are 100-feet wide and 1.8 miles long. The canal is built in the mid-1800’s as a major industrial transportation route. Historically was surrounded by heavy industries such as gas plants, concrete plants, industrial plants, chemical plants, tanneries and paper mills.
In the laboratory, identification of an unknown bacterium is often necessary. In the lab, a random sample consisting of three different bacteria was selected. The sample contained one gram-positive, one gram-negative paracolon, and one gram-negative coliform. The purpose of the experiment is to identify each of the three species that the mixture contained. After receiving an unknown mixture, the sample was streaked for isolation onto TSA, blood agar, and MacConkey plates.

The second ½ of the organism was used for gram staining. The gram stain method was performed on the unknown organism per lab manual page 42 and two gram stain reactions were identified. Organism B was gram positive cocci in grape like clusters. Because organism B was positive I could eliminate Escherichia coli, Enterobacter aerogenes and Proteus vulgaris because these bacteria would be rod shapes. Organism A was gram negative pink rod shaped and because of gram positive morphology I could eliminate Staphylococcus aureus, Streptococcus lactis and Bacillus subtilis.

The sterile cotton swab was inserted in the S. epidermidis culture and twirled around to obtain a specimen.
