Culture Media The enrichment medium (Table 1) is a variety of culture media that makes the environment conducive for the promotion of growth of a particular organism whilst not containing inhibitory substances to prevent growth of competitors. The BHI is a general-purpose enrichment medium for the cultivation of most organisms (Sigmaaldritch Catalog, 2012). It can further be mixed with antibiotics to allow it to be selective in nature. The addition of 0.1% agarose reduces the flow of oxygen and solidifies the medium hence favorable for the growth of anaerobic organisms (Atlas, 2010). The sterilization parameter can be used to assess the optimum temperature for the sterilization of media without sacrificing the nutrients it contains, variation of temperature can be done and checking for sterility may be via use of biological indicators containing Bacillus stearothermophilus spores for killing efficacy. Another parameter is the physical parameter wherein checking of the state of the medium whether it contains bubbles, cracks, agar height and alike. Also, noting for the pH of the medium is part of the physical aspect. The microbiological parameter deals with growth of organisms using CLSI standard guidelines via use of 0.5 McFarland Standard that should give atleast 1x107-1x108 CFU/mL (0.08-0.1 Absorbance at 625nm). Dilutions of 1:10 and 1:100 can be used for quantitation of colonies in plated media. The econometric parameter deals with the relationship Absolute Growth Index (AGI) and the Relative Growth Index (RGI). These parameters can be used for assessment of media by comparing the test plate to that of the control plate RGI can be computed by getting the ratio of the AGI of the test medium and the AGI of the control medium multiplied by 100. Lactobacillus lactis was inoculated in various media containing different vegetal source instead of beef extracts and was compared to the MRS culture medium as standard. These were incubated at 37oC for 72 hours wherein the growth was recorded every 24 hours using an undiluted, 10-4, 10-5 and 10-7 CFU/ml. The results of cultivation showed that the alternative test media using seed powders demonstrated better growth of colonies than the Man Rogosa Sharpe (MRS) cultre medium. Orange juice was used as an enrichment means for the enumeration of Alicyclobacillus acidoterrestis.
