Tacrolimus is specific T-cell inhibitor and potent immunosuppressant agent [no]. Tacrolimus (FK506 or Fujimycin) is a 23-membered polyketide macrolide with immunosuppressant activity that was discovered in culture broth of the soil bacterium Streptomyces tsukubaensis in 1984 . The immunosuppressant tacrolimus was initially discovered in fermentation broth of Streptomyces tsukubaensis, which was isolated from Japan soil of the Tsukuba region. Most of the initial studies performed with streptomyces tsukubaensis had been focused on strain improvement and development of fermentation procedures at industrial level . In initial studies Fujisawa scientists isolate a tacrolimus producing strain of streptomyces tsukubeansis, no 9993 and established
citri (Xcc), was provided by the Bacterial Citrus Canker Collection of National Institute of Genetic Engineering and Biotechnology (NIGEB). The bacterium was cultured on YPGA (3 g/l yeast extract, 5 g/l peptone, 7 g/l glucose solidified with 15 g/l agar) at 28 °C for 48 h. A single colony was subcultured in YP medium (3 g/l yeast extract and 5 g/l peptone) on a rotary shaker at 180 rpm at 28 °C till OD600 = 0.3, i.e., 5 × 108 cfu/ml. To prepare the inoculum, the culture was centrifuged for 10 min at 5000 ×g and the pellet was suspended with dH2O to reach a concentration of 1 × 105 cfu/ml. For inoculation, host plant leaves were surface-sterilized with ethanol and the inoculum (500 µl of 1 × 105 cfu/ml) was injected into the parenchymal space on the abaxial leaf side. After stirring/mixing the colonies for about 60 seconds we should look for the appearance of viscous nature and in that case the loop should be raised very slowly to see the formation of stringiness indicating that it is a gram negative bacterium [Figure 3]. Gram positive bacteria don’t form viscous emulsion and are negative for string like formation. Isolated bacterium was found to be negative for string test and was resistant to vancomycin 5 µg and sensitive to colistin 10 µg. Biochemically the bacterium was observed to be catalase positive, Indole, urease and citrate negative and showed no reaction/inert in tripple sugar iron agar (TSI) indicating that the organism has neither the ability to metabolise sugar nor the peptone. Antimicrobial susceptibility pattern of the isolated bacterium revealed sensitivity to imipenem, ciprofloxacin, ofloxacin, amikacin, piperacillin-tazobactum and cephalosporins including ceftriaxone, cefotaxime, ceftazidime, cefipime, cefipime-tazobactim and resistant to trimethoprim-sulfamethox azole.
SYNTHESIS AND CHARACTERIZATION OF Mg-Cu-Zn FERRITES NANOPARTICLE BY CO-PRECIPITATION METHOD OXALATE AS PRECURSOR M.SATYA VANI Lecturer in Physics D.N.R.College, Bhimavaram Abstract Ferrite nanoparticles of basic composition Mg0.40,Cu0.25,Zn0.50Fe2O4 were synthesized through co-precipitation wet synthetic method by using oxalate as precursor at 90°C then filtered and washed with distilled water. After drying, heat treatment was carried out for 3 hours at 600°C and the resulting compounds were characterized for structural properties using X-ray diffraction [XRD], scanning electron microscopy, transmission electron microscopy, and Fourier transform infrared spectroscopy [FT-IR]. The XRD result shows that all prepared samples crystallite size are in the range of 30 nm to 60 nm and lattice constant in the range of 8.20 to 8.40 nm. Fourier Transform Infra Red Spectroscopy results clearly indicate the Mg-Cu-Zn nano Ferrite are synthesized. Differential scanning calorimeter graphs shows the phase formation of all the samples.
ASSESSMENT OF CADMIUM AND LEAD CONTENTS IN TISSUES OF TILAPIA AND CAT FISH SPECIES SOLD IN NSUKKA, NIGERIA Abstract The levels of cadmium and lead in the tissues of dried Clarias gariepinus and Oreochromis niloticus fish sold in Nsukka were investigated using Atomic Absorption Spectrometer. The study revealed mean Cd levels of 0.0015 and 0.0024µgg-1 for bones, gills and flesh respectively with a grand mean level of 0.0018µgg-1 in dried Clarias gariepinus. The average Pb concentrations of 0.0384, 0.0752 and 0.0299 µgg-1 for bones, gills and flesh respectively of the same dried Clarias gariepinus were also obtained with a grand mean level of 0.0478µgg-1. This study further revealed mean Cd concentration of the tissues of dried Oreochromis niloticus
