Lab 7 Report: Molecular Methods - Group 5 Abstract. This lab report outlines the procedures and findings of an RT-qPCR experiment conducted on extracted DNA from E. coli K-12 samples. The objective was to assess microbial dynamics and investigate the efficacy of molecular methods in detecting specific pathogens. The experiment involved DNA extraction using silica columns and various buffers, followed by qPCR setup and analysis. Very precise procedures and protocols for biosafety measures and quality control, including technical replicates and dilutions, were used throughout the experiment. Results revealed concentrations of extracted DNA and provided insights into microbial dynamics. Despite challenges such as potential contamination during sample handling, the experiment showed the versatility of qPCR in targeted microbial detection and highlighted the importance of methodological rigor in molecular biology research. It's important to mention that the last wells for the sample in Table 1 were not run due to not having enough master mix which is why its average concentration is not available, which shows how specific this lab is with handling materials and mixtures. Overall, this study contributes to our understanding of nucleic acid extraction techniques and their applications in microbial analysis, with implications for …show more content…

The master mix is used to ensure components other than the primers and probe are present for successful qPCR. Forward primer: the forward primer is used to bind to the template DNA. Reverse primer: the reverse primer is used to bind to the non-template DNA strand. FAM probe: used to target a specific target sequence. Fluorescent dyes attach to the probe, which then target a portion of the DNA target sequence. Only this sequence will amplify. Water: used to get to the final reaction
