Yakult was purchased from well known market in Vellore, Tamil Nadu, India. Ingredients of Yakult: Skimmed Milk Powder, Sugar, Glucose, Natural and Natural Identical Flavour, Water and 6.5 billion Lactobacillus casei strain Shirota. MRS agar and MRS broth were collected from Hi Media Laboratory, Mumbai, India. MRS agar contains peptone (1.0%), egg extract (0.8%), yeast extract (0.4%), glucose (2.0%), sodium acetate trihydrate (0.5%), polysorbate 80 (0.1%), dipotassium hydrogen phosphate (0.2%), triammonium citrate (0.2%), magnesium sulfate heptahydrate (0.02%), magnesium sulfate tetra hydrate (0.005%), agar (1.0%). pH adjusted to 6.2 at 25 ºC. The yeast and meat extracts and peptone provide sources of carbon, nitrogen and vitamins for general bacterial growth. The yeast extract also contains vitamins and amino acids specifically required by Lactobacilli. Polysorbate 80 is a surfactant which assists in nutrient uptake by Lactobacilli.
