MRS agar and MRS broth were collected from Hi Media Laboratory, Mumbai, India. MRS agar contains peptone (1.0%), egg extract (0.8%), yeast extract (0.4%), glucose (2.0%), sodium acetate trihydrate (0.5%), polysorbate 80 (0.1%), dipotassium hydrogen phosphate (0.2%), triammonium citrate (0.2%), magnesium sulfate heptahydrate (0.02%), magnesium sulfate tetra hydrate (0.005%), agar (1.0%). pH adjusted to 6.2 at 25 ºC. The yeast and meat extracts and peptone provide sources of carbon, nitrogen and vitamins for general bacterial growth. The yeast extract also contains vitamins and amino acids specifically required by Lactobacilli. Polysorbate 80 is a surfactant which assists in nutrient uptake by Lactobacilli. First, one measured and then added seven grams of Fleischmann 's Rapid-Rise yeast to a bottle containing two hundred and fifty milliliters of warm distilled water for the purposes of creating the standard yeast solution that was used in the fermentation experiment. Next, the standard yeast solution was added to each of the four fermentation flasks. After swirling the bottle, one and a half grams of the fermentation substrate was added into a beaker with fifteen milliliters of yeast suspension. Gram’s stain was originally devised by histologist Hans Christian Gram in 1884. Gram-positive bacteria stain purple, while Gram-negative bacteria stain pink when subjected to Gram staining.
Strengths: As a student one of my strengths is organization. I can say I can keep my school materials and notebooks organized in a way that I and others can comprehend. The reason I can say I am organized is because, I have hardly ever had in the past or present any issues with trying to find homework or assignments because I always kept my materials for each class organized and in a place I could easily find.
