Abstract The purpose of this experiment is to test for mitochondrial activity by isolating different organelles using the differential centrifugation process. Studying mitochondria is extremely important because they control the death and life of the cell by regulating the apoptotic signals (Frezza et al 2007). Also they are responsible for the metabolic reactions (aerobic respiration) and the production of ATP (Frezza et al 2007). Three hypotheses were formed based on my knowledge. First, it was hypothesized that test tube "A", the control, would not show any red concentration, test tube "B" which contains supernatant II would show the most red concentration and test tube "C" which contains sediment II would only show a little red concentration. The second hypothesis states that the raw corn kernels would have mitochondrial activity while the boiled corn kernels would not. The last hypothesis interprets that the "gunk" and sediment I will both contain starch granules. It was only expected to find mitochondrial activity in Supernatant II. Unfortunately, after performing this experiment, we were not able to support this hypothesis and come up with a conclusion. A possible explanation for why there was no mitochondrial activity observed in the boiled corn kernels might be because increasing the temperature would disintegrate dehydrogenases and thus they wouldn't function as efficient or they …show more content…
For instance, we could not conclude that mitochondrial activity is present in Supernatant II. However, our experiment showed that the boiled corn kernels did not undergo any mitochondrial activity while the raw corn kernels did. This might indicate that raising the temperature might have an effect on the function of dehydrogenase. Moreover, our found that starch granules are present in both sediment I and the “gunk”. Indeed, some parts of this experiment were not successful because the procedure was not followed
Fermentation test is used to determine if unknown #398 uses any oxygen to ferment carbohydrates and acids. Oxidation tests were used to determine if unknown #398 metabolizes carbohydrates and acids by cellular respiration. Both tests are observed by inoculation of unknown #398 into 3 sugar broths: lactose, glucose, and mannitol and 1 citrate (Citric acid) slant. Fifth test, Hydrolytic and Degradative reactions is used to determine if unknown #398 contains enzyme, amylase that hydrolyzes starch after streaking on a starch plate. Next test, inoculation of a urea broth and is used to determine if unknown #398 contains urease that hydrolyzes urea.

During this experiment, mitochondria were isolated from 20.2 grams of cauliflower using extraction buffer, filtration through Miracloth, and centrifusion. Twelve samples containing various volumes of mitochondrial suspension, assay buffer, DCIP, sodium azide, and citric acid cycle intermediates were prepared to be read by a spectrophotometer. The inclusion of the dye DCIP allowed for the absorbance of the reactions between the mitochondrial suspension and the TCA cycle intermediates succinate, malonate, and oxalate to be measured, as DCIP turns from blue to colorless as the activity of succinate dehydrogenase increases. Experimental Findings Increasing the number of mitochondria in the reaction did increase the reduction of DCIP relative to the amount of mitochondrial suspension present.
Starch solution is then placed into the test tube at a quantity of 5 mL. 5 drops of Lugol’s Iodine solution is added to the test tube. If the color changes, then it is known that starches are present in the solution. Proteins are next tested. In order to do this, 5 mL of gelatin solution is added to the test tube. 10 drops of Biuret’s reagent are added to test for protein.

For this assignment, a summary was to be written to explain the scientific article that I chose and relate that article to cellular respiration. The title of the article is: Revisiting the Relationship Between Exercise Heart Rate and Music Tempo Preference. The main purpose of this article related to cellular respiration is that music helps make a positive effect on a person, which increases the need to exercise which, in turn, raises the heart rate so this correlates to the purpose of the lab report. Preference is important in music so it can either be slow music or up-tempo music which would either increase or decrease the need to exercise more for an increase or decrease of heart rate. The iodine test determines the presence of starch in biological materials. It is predicted that, if starch is not present, the solution with iodine remains yellow. However, if starch is present the solution with iodine becomes a blue-black colour. Plants have starch as the storage polysaccharide (glucose units held together by glycosidic bonds) while animals have the equivalent of glycogen. In this experiment, the dark blue colour is visible because of the helical amylose and amylopectin reacting with iodine (Travers et al., 2002).

Introduction: Respiration encompasses cellular respiration and external respiration. In cellular respiration the vast majority of energy that most living organisms need are satisfied by the mitochondria. The main driving force behind the mitochondria producing the high yield of ATP would be due to oxygen gas. For external respiration, encompasses breathing, gas exchange in the alveoli, transportation of gases through the blood, the delivery of gases to the systemic tissues, and the gas exchange at the systemic capillaries.
The 3 concentrations of enzymes were 0.5 ml, 1.0 ml, and 2.0 ml of turnip extract, while the substrate consisted of 0.1ml, 0.2 ml, and 0.4 ml of hydrogen peroxide. In a separate tube, the control was made up of turnip extract and guaiacol, known as the color reagent. This was recorded the absorbance every 20 seconds for 3 minutes.
Eukaryotic cells contain many important organelles and without them the cell cannot function accurately. With organelles such as the nucleus which directs cell activity and contains DNA, ribosomes which make protein, the vacuole which is used for storage and in order for the cell to survive; the mitochondria. The mitochondria are often described as the energy powerhouse of the cell as organisms need energy to maintain homeostasis. The mitochondria are found in the cell cytoplasm and are double membrane enclosed organelles ‘which is best known for its critical function in energy production via oxidative phosphorylation, a pathway that generates many more ATP molecules per glucose molecules than glycolysis’ (John Wiley & sons, 2009) [2].
